Abstract:

This report contains the standard protocol used for fungal transformations using the Agrobacterium-mediated transformation (AMT) method. Users should be able to conduct AMT experiments using the following standard protocols. Recipes for media used for transformation are included in this role. A brief background to AMT and the different Agrobacteria strains are also described to help end users in selecting the correct strains for their research. Discussion includes considerations for AMT use in fungal research. Each fungal isolate may have its own peculiarities, and some tend to be recalcitrant. The following protocol has been used at ARL for the chromosomal integration of DNA into 16 different Ascomycete fungal strains.
